We have synthesized a 582,970-base pair Mycoplasma genitalium genome. This synthetic genome, named M. genitalium JCVI-1.0, contains all the genes of wild-type M. genitalium G37 except MG408, which was disrupted by an antibiotic marker to block pathogenicity and to allow for selection. To identify the genome as synthetic, we inserted "watermarks" at intergenic sites known to tolerate transposon insertions. Overlapping "cassettes" of 5 to 7 kilobases (kb), assembled from chemically synthesized oligonucleotides, were joined by in vitro recombination to produce intermediate (...) assemblies of approximately 24 kb, 72 kb ("1/8 genome"), and 144 kb ("1/4 genome"), which were all cloned as bacterial artificial chromosomes in Escherichia coli. Most of these intermediate clones were sequenced, and clones of all four 1/4 genomes with the correct sequence were identified. The complete synthetic genome was assembled by transformation-associated recombination cloning in the yeast Saccharomyces cerevisiae, then isolated and sequenced. A clone with the correct sequence was identified. The methods described here will be generally useful for constructing large DNA molecules from chemically synthesized pieces and also from combinations of natural and synthetic DNA segments. 10.1126/science.1151721. Are words like ‘woman’ or ‘man’ sex terms that we use to talk about biological features of individuals? Are they gender terms that we use to talk about non-biological features e.g. social roles? Contextualists answer both questions affirmatively, arguing that these terms concern biological or non-biological features depending on context. I argue that a recent version of contextualism from Jennifer Saul that Esa Diaz-Leon develops doesn't exhibit the right kind of flexibility to capture our theoretical intuitions or moral and political (...) practices concerning our uses of these words. I then float the view that terms like 'woman' or 'man' are polysemous, arguing that it makes better sense of the significance of some forms of criticisms of mainstream gender ideology. (shrink)

Analytical study of the social theories of Muslim philosophers belonged to 9th to 18th centuries.

Those who have studied the Athenian system of command in the fifth century have confined themselves almost entirely to the period after 440 B.C. They have raked over the evidence to discover signs of double representation of one tribe on the board of strategi, or of a supreme among the or of a chairman at least of the board of strategi. On the other hand little attention is paid to the progressive diminution of the military functions of the archon polemarchus (...) within the state and to the great problems created in external affairs by the Persian Wars with the formation of the Greek League and then of the Athenian Alliance. Yet these matters are vital to the evolution of the system of command which can be seen in operation after 440 B.C. In particular the decisive steps were probably taken in 480–466 B.C., when Athens' national system of command had first to be integrated into a command-system of combined forces headed by Sparta in the Persian Wars and then adapted to take over the command of combined forces in continuous warfare against Persia. In this article I try to study the whole field and to avoid applying to the early part of the period the theories which have been evolved hitherto with special reference to Pericles in 440–428 B.C. Upon doing something generous for someone with whom you are close, some kind of reciprocity may be appropriate. But it often seems wrong to actually request reciprocity. This chapter explores the wrongness in making these requests, and why they can nevertheless appear appropriate. After considering several explanations for the wrongness at issue (involving, e.g. distinguishing oughts from obligation, the suberogatory, imperfect duties, and gift-giving norms), a novel proposal is advanced. The requests are disrespectful; they express that their agent insufficiently trusts (...) the hearer to recognize their own reasons to reciprocate, and the close relationship between them morally requires this kind of trust. This proposal is articulated and situated in the recent discussion of the normativity of requests. The chapter concludes by explaining how these requests may appear appropriate. Agents in these relationships often have standing to make requests, though the standing is lacking in these particular cases. (shrink)

It was a characteristic of Macedonian custom that a name was used in a special and in a general sense. For example, ‘Foot-Companions’ was the name of a Bodyguard of Philip and also of the men of the Phalanx-Brigades from Lower Macedonia, and ‘Hypaspists’ was the name of Infantry-Guardsmen of Alexander and also of the men of three Hypaspist Phalanx-Brigades. Geographical names were repeated: there were at least two regions and two cities called ‘Emathia’, two or three regions called ‘Doberus’, (...) four cites called Philippi or Philippopolis, and Alexandrias galore. To a modern mind this makes for confusion. That is certainly the case with the names ‘Macedonia’ and ‘Macedones’, which I shall treat separately in this article. (shrink)

Theoretical determinism, as it is usually ascribed to Laplace, is neither verifiable nor falsifiable and has therefore no real content. It is not the same as predictability of actually observable phenomena. On the other hand, predictability is not an abstract principle; rather it is true to a certain degree, depending on the phenomena considered. It can be discussed only by examining the scientific state of affairs. This is done in some detail for classical statistical mechanics. Much of a recently published (...) debate on determinism (Amsterdamski et al. 1990) is thereby obviated. (shrink)

A critical survey of various positions on the nature, use, possession, and analysis of normative concepts. We frame our treatment around G.E. Moore’s Open Question Argument, and the ways metaethicists have responded by departing from a Classical Theory of concepts. In addition to the Classical Theory, we discuss synthetic naturalism, noncognitivism (expressivist and inferentialist), prototype theory, network theory, and empirical linguistic approaches. Although written for a general philosophical audience, we attempt to provide a new perspective and highlight some underappreciated problems (...) about normative concepts. (shrink)

In this article I try to give a precise meaning to the political terms which are used in the sources, and to use those terms for an understanding of political developments. I take these developments in their historical order, and at the end of each period I assess the value of the evidence. I finish with reflection on the contributory system and the hegemony of the Thebans.

Ethicists struggle to take reductive views seriously. They also have trouble conceiving of some supervenience failures. Understanding why provides further evidence for a kind of hybrid view of normative concept use.

NG van Kampen is a well-known theoretical physicist who has had a long and distinguished career. His research covers scattering theory, plasma physics, statistical mechanics, and various mathematical aspects of physics. In addition to his scientific work, he has written a number of papers about more general aspects of science. An indefatigable fighter for intellectual honesty and clarity, he has pointed out repeatedly that the fundamental ideas of physics have been needlessly obscured. As those papers appeared in various journals, partly (...) in Dutch, it was felt that it would be worthwhile to collect them (translating the Dutch material into English) and make them available to a larger audience. Arrian was better qualified to understand the nature and significance of ‘the pursuit’ in Macedonian warfare than any modern scholar. He had himself fought and commanded in a very similar kind of warfare, and he was keenly interested in the study of military tactics. He was also better informed about the pursuits which Alexander had conducted, because he was able to use the accounts of Alexander's contemporaries, Ptolemy and Aristobulus. Anyone today who wishes to question the veracity of Arrian's reports (...) of these pursuits cannot do so of his own experience. He should therefore turn to comparable pursuits of recent times which have been reported accurately beyond any shadow of doubt. (shrink)

Anyone who reads the opening chapters of Thucydides’ history consecutively will soon find it difficult to follow the thread of the argument. If he turns to a summary of the subjects chapter by chapter, he will not be greatly enlightened. In this paper the question is asked: why did Thucydides arrange his subjects as he did? In Part I the conclusion is reached that in the arrangement of his subject-matter he was following a clear-cut system. In Part II the implications (...) of this conclusion are considered. (shrink)

The revolution in science and technology is significantly changing the character of production and work. One of the important aspects of this change is that labor becomes a scientifically organized process that makes increased demands on development of the human factor in production. The most productive scientific ideas, the most perfect technologies, when combined with production, may fail to produce the desired "yield" unless a sufficient number of well-trained personnel, or of the required organizational structures corresponding to the level of (...) societal relationships, the habits of collective work, and human psychology, are available. Implementation of a truly scientific organization of work presumes as a condition that a considerable level of development will already have been achieved in socialist labor discipline, as well as habits of collectivism and ethical motivations for work, and not merely occupational skills and qualities. More precisely, the list of qualities that, while they may not be occupational in the strict sense are nonetheless necessary features of the person employed in socialist society, includes general cultural and moral development, social activity, and political consciousness. The transformation of work into a scientifically organized process presumes an increasing demand for a highly skilled labor force with comprehensive development. Moreover, the development of the labor force and the rise in its skills should run ahead of the development of the means of production. It is precisely this that determines the need for conscious management of the development of the human being. For comprehensive development of the labor force to run ahead of the means of production requires considerable outlays for occupational and general cultural training of the direct participants in production. A worker of a new type, with a higher level of general and occupational education, will emerge in the course of implementation of a complex of economic, social, and scientific-technological measures.
